I'm looking at a Lee Enfield No1 MkIII*, but it is missing a screw at the end of the cocking piece.

Is it safe to fire without this screw? Where could you get a replacement?? Any info would be appreciated.

It's the firing pin locking screw, you could fire without it, but the firing pin could shift and cause light primer hits or puncture the primers. I would recommend not firing until the screw is on place.

As for replacement check, marstar or numrich in the states if you need multiple parts.

It's in stock at marstar http://marstar.ca/dynamic/product.jsp?productid=78400

A CGN member on here fixs up alot of these rifles .... goes by louthepou and he lives here in Ont .
Maybe he can help you out.

You can fire safely without it, the worst than can happen is soft primer strikes, the pin will not punch the primer as the pin is stopped by the collar on the pin strikeing the back of the bolthead so unless you already have excessive firing pin protrusion this will not be an issue.
