Remove barrel from a Ruger Vaquero

They're right hand twist threads but you need to be very careful. Some of those barrels are extremely tight and if you aren't careful, you might damage both the frame and the barrel.

Take it to someone who knows what they're doing.

I've seen revolvers, both double action and single action with bent frames and scored barrels, which were no longer usable.
 
Well you do not want to remove the grips and place a large Crescent wrench on the grip frame. You want to use a wrench that engages the revolver frame as close to the barrel as possible. Revolver frames are easily distorted/twisted.
 
If it's a 44-40 they were notorious for cheaping out using .44 mag barrels .430" diameter while leaving the cylinders .427 Stupid move IMO the cure is having the cylinder opened up to match the groove. Same smith I mentioned prior can do that then they really shoot. Starline and Win brass are thinnest allowing larger bullets but you will also need cowboy dies /RCBS Rem brass is the thickest and NFG for this problem.
