The major difference between the Mark III's and the 22/25's are the grip angle. The Mark III's have an well-angled grip and 22/45's have a more traditional vertical grip to them. The 22/45's are supposed to replicated the feel of a 1911. There are also slight differences in styling and the mags are different.

There are always pros and cons about different barrel length, general speaking, a longer barrel has longer sight radius and is more accurate but it also heavier and not quite easy for rapid fire. I found the 5.5 inches bull barrel one on the MarkII/III is very comfort to shoot and I like the grip angle too.
