The biggest problem your going to have shooting from a tree stand isn't wether you're shooting downhill, but the amount of height your scope crosshairs are over the bore and the range you're sighted for.
20 yards or so isn't a problem with body shots on deer and larger animals unless you're going for a neck or head shot.
When you're very close, the distance between the center of the bore and the crosshairs can be critical.
In many cases, especially with 50mm objective lenses, the crosshair can be 3-4cm above the bore. Therefore the point of impact can be up to 4cm low.

Always shoot down hill, it is far easier than walking up hill.
