I'm looking to buy a single action revolver with a loading port in the near future. So far I know of Ruger that makes them. And that company through Marstar. Are there any others?

Marstar carries the Pietta line in .357mag and .45colt. Here's mine in .45colt. They are really nice guns, fit and finish is superb. Price is right too.

I also have this little .22lr/.22mag single action. Decent revolver made in Germany (Model HS21). They can be found quite cheap, aalot cheaper than the Ruger single six's.

All single action revolvers have loading ports.

If they didn't have this port, the cylinder would have to be removed to reload the gun.

Actually you're a bit wrong on this one - the Smith & Wesson 1875 Schofield Single Action revolver is a break-top (ie, no loading port). Uberti currently makes repros and S&W has re-introduced the real deal in .45 Schofield (essentially a shorter verson of the .45 Long Colt).
