If you want more velocity than the .243 Winchester, the 6mm Remington or .240 Weatherby, or one of the legions of old 6mm wildcats are probably better choices. The .243 WSSM gives you about 10% increase over the .243 (comparable to the 6mm), but brass is harder to find and the cartridge is trickier to develop for--brass is very thick around the neck, which can create problems with seating and chambering, and pressure problems tend to come up FAST, with few early warning signs.
If you're going the custom route, maybe check out the 6mm-06 wildcat. You can make brass easily from .25-06 factory, and it performs on par with the .240 Wby.
