In the past I've owned and shot .455 in Webley Mk. III and Mk. IV revolvers, also a Colt New Service and a S&W Hand Ejector.
I used a Lyman 454190 RNFP mold mostly (which is still listed as in production), and also an old Lyman 454424 (no longer produced) and sized my bullets to .454".
I used both Webley Mk. I and Mk. II length brass, including old 'balloon head' Dominion .455, newer .45 Long Colt brass modified to Mk. I specs, and newer Starline Mk. II type .455 brass.
The above bullets shot well out of all my revolvers, but the 454190 was my favorite.
I never had a cylinder modified for 45ACP, and my understanding is that moon clips are required to be used with that modification but are fairly readily available. If your .454 bullets in 45ACP brass aren't too big in diameter, and will load easily into that cylinder, then all's good.
As you have both cylinders you might want to see if shortened .45 Long Colt brass will fit the ACP cylinder and still leave sufficient clearance to rotate easily with no drag without needing rim thinning.
You also have that unshaved cylinder, and .455 Mk. II brass is usually quite available, so you can have the best of both worlds, but the 454190 mold would be the one for me.
