Bullets meant for muzzleloaders such as the ones that you mention are generally going to be under .500.
The bullets that you want for loading cartridges should be about 2 thou larger than bore diameter. On old 50's that will vary but could be as large as .510 or more. Slugging the bore will tell you for sure.
As for charges I go with what they were made for in things like this and would start with enough 1F or 2F powder to give about 1/8 to 1/4 inch compression when seating the bullet. There is a sticky about smokeless loads in antique rifles if you want to go that way too though.
The last one i made bullets for slugged at .512", the same size as my .50 Sharps are, bullets are usually very short and around 260 grs., I think some bores go to .515".
