I'm going to bed the action on a 527. I'm going to use Devcon 10110 and have also got some adjustable pillars from brownells. I would like to put some threaded rod in the action screw holes as guides but can't seem to find what kind of thread pattern it has, if anyone knows please tell me. The second question I have concerns the pillars, do I need them if I'm glassbedding, what will I gain?

Pillars are always a good idea.

As far as guide bolts, WHY???? Just put lots of release agent on your bedding bolts and use them.

Before you pour your resin, make sure the pillars are the exact length they need to be. Once they are glassed in place, the only way to shorten them easily is with a milling machine.

When I bed an action, I assemble the rifle with everything in place, all gooped up with release agent of course. I also use an old playing card under the barrel at the fore end tip. This is just a quick way to fully float the barrel without having to resort to rasping out the barrel channel.

Depending on how much of your action you intend to bed, you may want to remove the trigger group.

I find with the CZ527 that it really responds well to a full receiver glass bedding, rather than just the front and rear. It only takes a few minutes to clean up any excess bedding material that you feel will cause issues.
