I recently bought a lightly used singlesix from the original owner and when it arrived I noticed that the drag marks on the cylinder were a off center by a fair bit....I've got other revolvers that are a little off but this one is the worst I've seen.....it seems to engage the lugs ok however so I was not to worried about it.
When I finally got to the range I noticed that the base pin would not stay in place for more then a few shots and would slowly creep forward till the cylinder was binding a bit.... which is when I noticed it.
I have heard of base pins moving on heavy recoiling revolvers but never thought it was an issue on a .22 and in this case as you can see in the video below just dry firing the revolver twice is enough to dislodge the pin!
I have not really looked at the spring loaded plunger part that holds it in but I'm thinking I should take it apart and see what's up but I thought maybe I'd post here and see if anyone else has had similar issues before?

The name of the part you are referring to is called a cylinder stop.. As long as your cylinder locks up when you #### it I would not be tooooo concerned. The stop should protude through the bottom of the frame. Take the cylinder out and try to move the stop into the frame, it should move freely. There may be a burr which is causing the cylinder drag mark to be offset.

The cylinder latch looks has a bit of a burr from rubbing against the stop but mostly it's just not centered when it is fully engaged.
Here's a picture of my single six compared to a black hawk that is centered,

Looking some more at the base pin and the base pin latch it seems like the latch is not sliding over enough to fully engage the groove in the pin...it only takes a hair of movement of the latch to disengage the pin.
 
Well I took the latch assembly apart (a cheap bit from a "security" set of bits I had worked great for the tricky side:))

Turns out there is a burr inside the frame where it was machined for the latch...I think this little flap of metal was not letting the latch fully seat into the groove on the pin!

Cleaned it up and reassembled and it seems to have cured the Base pin issue!

I also smoothed out the edge of the cylinder latch.:)
I didn't get to shoot it allot on the first outing because it was getting dark but the accuracy seemed poor hopefully this was due to the base pin being loose!!
Looking forward to the next range trip!
