The half moon clips are for the version of the N-frame that is chambered for the .45ACP cartridge. Being a rimless cartridge, the clips allow the round to be fired & extracted from the cylinder. In the model 27, as the .357Mag & 38 SPL are rimmed, half moon clips are not required.

I'm not sure I'd have the cylinder done though, unless your building an IDPA or IPSC gun. Maybe buy a second cylinder and have it machined and fitted?
Or go for a 625 or 25-2 in .45ACP that's ready for clips already. The loading will also be easier on those ones with the bigger chamber diameter, too.Sell that one to a collector and buy a new one with the cylinder mods.
