I have been shooting cap and ball revolvers for a while now and have used the spout on the powder flask to measure the powder by volume. I recently bought an 1873 SAA cartridge revolver in .45 Colt and will be reloading for it using a Lee 4 hole turret press kit which comes with a powder scale which obviously measures the weight rather than the volume. I currently have Goex FFFG powder on hand but the Goex reloading data recommends 40-42 grains (powder charge measure setting) of FFG for the .45 Colt cartridge depending on the weight of the bullet.
"Powder charge measure setting" sounds like a volume measurement to me and I could come close using a combination of the spouts that I have.
Can I safely use FFFG in this cartridge instead of the FFG as recommended or should I reduce the charge and use a wad to make up the space below the bullet.

I use Goex 3F in all of my .45 Colt loads... I load by volume and not weight. I use a shortened .45 Colt case trimmed to hold 35 grains by volume as a measure. Soldering a piece of heavy copper wire to the side of the case makes a good handle for convenience.
 
I load .44-40 and all my ammunition by weight because accurate volume measurement depends on having a consistent density and I prefer to have my ammunition as consistent as possible. Wither you need a measurement to +/- 0.05 grains is another matter when using black powder. I would consider it necessary for smokeless powder..
 
Black can be measured by volume or weight I use weight for all my target loads volume for plinking I have a Hornady black powder measure and just drop by volume when I want to shoot a bunch. Pyrodex they say only by volume on the can as it has a lower specific gravity .
