Can't help with titegroup, honestly I wouldn't use it in an antique but that's just my opinion
For trail boss, do exactly what the manufacture recommends, fill a case to the level a seated bullet will sit, weigh out that charge and that's your maximim. minus 10% and you're good to go

I was using 4gr trailboss in a .45 acp case with 255/65 gr. Recoil was around half of standard .45 acp ball. not sure of OAL but i could check, i was crimping in the provided groove.
4.2-5 gr trailboss is prob the max with 3.5 gr being light. Thought ive tested please do additional research and take with a grain of salt.

Are you using 455 mark2 brass or 45acp/ar? Also what type of bullet are you using? Semiwadcutter? There are lots of variables involved but the big three are: bullet weight, powder charge and seating depth.
Made 455 mark 1(455 eley) brass from cowboy 45 special. Cut to length, ream out case necks. Turn down the rims.
Probably use old Keith 255 grain 454" mold.
3.5 g of Titegroup shouldn't make much pressure I'm guessing.

Ah a Keith I have some of these so I can make some calculations, what length did you make the cases? I've used unmodified 45 cowboy in a Belgian 450 Adams
Using the 22.5mm case length on Wikipedia and seated at the canalour ~9mm deep, 3.5 grains should give velocity in the mid 600s and a pressure of ~12000psi, that's technically within limits but pretty close for my comfort,
Accordingly if you crimp it in the lube groove at ~6mm seating depth and using 4grains should get the same velocity but only a little over 10000psi, all that being said these are just models and tightgroup is a very fast powder for such a heavy bullet with alot of inertia, consider something like WST powder in the future, it has basically the same burn rate and powder weight as bullseye which there is plenty of info written about for Webleys
