That's a tough question, but I think I have to go with the Sig X5 Level 1(Gen1) I got from Harry C a few years back. I like it so much, I actually traded my nickel Python recently for a new X5 Supermatch, as last buy/trade before they shut us down. I figured another X5 was in order.
The X5 L1 is just so nice to shoot, and shoot well. Makes me look better then I am. The quality of the build is superb. Pictured is the X5 L1 and Gold Cup National Match.

Interesting I didn't know Ruger ever made a 45 win mag cylinder. Always learning new things. With a shorter barrel that Blackhawk would be a handy package in the back country, with the right paperwork or course. You'll probably remember in the 80s, North American Arm made a single action 450 Express, that came with a 45 win mag cylinder.

I have to update this post. My new favorite is still an X5, but the new Supermatch that I just got. A couple of small details that I really like, one of which is the heavy takedown lever which is just *made* to index my left thumb on. I also really like the flat rather then curved trigger. First gun I've owned with it and I quite like it! I don't know that it shoots any better then the L1, but I sure like the grips (Hogue G10s) and those two small details I mentioned. New favorite!
The sharp eyed will note that I painted the front sight bright orange. I just can't see a black front sight anymore, they all get painted now.

That's a beauty TC. I had one awhile back and agree it is a phenomenal gun. Curious to see your comparison of the 2 generations. Some guys have suggested the Gen 1 are superior to the Gen 2, but I've had a few of each over the years and they are both exceptional. I appreciate the classic Gen 1 lines and the additional features of the Gen 2 that you noted. With the optic mounted, the Gen 2 is even more of a laser. Although I think they are now making rear sight optic mounts that also work on the Gen 1.

My favorite to shoot is a Springfield Mil Spec in .45. I bought it used about 15 years ago. The edges were dehorned from factory, it had a commander style hammer and trigger job done to it when I bought it. I swapped a homemade rat tail grip safety onto it, used it for my shade tree gunsmithing attempts, and never looked back.
Despite my modifications it is the best shooting pistol I've ever held. I'd never sell it. That being said, nobody would give me 10 dollars for it after what I've done to it.
I'll give you $10 for it, on the down low. Just meet me in Surrey or at the corner of Jane and Finch.
Two problems with that: it's registered, and the price is 5k if you want me to go to Surrey!
